RESUMO

Malignant peripheral nerve sheath tumor (MPNST) is a highly aggressive and recurrent soft tissue sarcoma. It most commonly occurs secondary to neurofibromatosis type I, and it has a 5-year survival rate of only 8-13%. To better study the tumor heterogeneity of MPNST and to develop diverse treatment options, more tumor-derived cell lines are needed to obtain richer biological information. Here, we established a primary cell line of relapsed MPNST RsNF cells derived from a patient diagnosed with NF1 and detected the presence of NF1 mutations and SUZ12 somatic mutations through whole-exome sequencing(WES). Through tumor molecular marker targeted sequencing and single-cell transcriptome sequencing, it was found that chromosome 7 copy number variation (CNV) was gained in this cell line, and ZNF804B, EGFR, etc., were overexpressed on chromosome 7. Therefore, RsNF cells can be used as a useful tool in NF1-associated MPNST genomic amplification studies and to develop new therapeutic strategies.

Mulberry zonate leaf spot disease (MZLSD) is an important fungal disease of mulberry trees, which seriously affects the productivity and quality of mulberry leaves. MZLSD has been widely reported in sericultural production areas in Guangxi, China, in recent years. In this study, the causal agent of MZLSD was isolated from symptomatic samples and identified as Gonatophragmium mori (Acrospermaceae) based on morphological characterization and molecular analyses using nucleotide sequences of the internal transcribed spacer (ITS) and large subunit ribosomal DNA (LSU rDNA). Pathogenicity tests confirmed that G. mori is the pathogen responsible for MZLSD. Furthermore, we isolated antagonistic endophytic bacteria (AEB) from healthy mulberry leaves. Plate confrontation experiments showed that the lipopeptide crude extracts (LPCE) of three endophytic bacteria can inhibit the growth of G. mori, and the diameter of the antibacterial circle reaches more than 60 mm when their concentration of LPCE is 200 mg/ml. Light microscopy and scanning electron microscopy revealed that LPCE caused drastic changes in mycelial morphology. Fluorescence microscopy and transmission electron microscopy showed that the LPCE-induced apoptosis-like cell death in G. mori hyphae. Finally, based on morphological and molecular features, we identified the three isolates as Bacillus subtilis DS07, B. subtilis DS32, and B. velezensis Q6, respectively. To our knowledge, this is the first time to identify G. mori by combining characterization and molecular analyses, and we provide timely information about the use of biocontrol agents for suppression of G. mori.

OBJECTIVE: To investigate the effect of contralateral controlled functional electrical stimulation (CCFES) combined with mirror therapy on motor function and negative mood in stroke patients. METHODS: Medical records of 94 stroke patients in Baoji Central Hospital admitted from April 2020 to October 2022 were retrospectively analyzed. Among them, 45 patients receiving routine rehabilitation training combined with mirror therapy were included in a control group, and 49 patients receiving CCFES combined with mirror therapy were in an observation group. Observation indexes included changes in Fugl-Meyer Assessment (FMA), Berg Balance Scale (BBS), Hamilton Anxiety Rating Scale (HAMA), Hamilton Depression Rating Scale (HAMD), Stroke Specific Quality of Life Scale (SS-QoL) score, and Barthel Index score before and after treatment. Patients with HAMA score >14 and HAMD score ≥20 after the treatment were included in a negative mood group, and logistics regression was used to analyze the risk factors for negative mood. RESULTS: The observation group had a significantly higher overall response rate after treatment compared to the control group. In addition, the observation group exhibited significantly higher scores in the FMA and BBS after treatment, indicating better physical function (P<0.001). Furthermore, the observation group showed lower HAMA and HAMD scores after treatment, suggesting reduced anxiety and depression levels (P<0.001). The quality-of-life scores measured by the SS-QoL and the Barthel Index score were both increased in the observation group after treatment, indicating better overall well-being and functional independence (P<0.001). Logistic regression analysis revealed that age, post-treatment SS-QoL scores, and post-treatment Barthel Index were identified as influencing factors for the onset of adverse emotions in patients (P<0.05). CONCLUSION: CCFES plus mirror therapy can effectively ameliorate limb function and lessen anxiety and depression in stroke patients, exerting beneficial effects on rehabilitation.

PURPOSE: In this study, we investigated the effects of mirror therapy (MT) combined with contralaterally controlled functional electrical stimulation (CCFES) on upper limb motor function, activities of daily life, and corticospinal excitability in post-stroke patients. METHODS: Sixty post-stroke patients were randomly divided into four groups: CCFES, MT, MT combined with CCFES, and control. All the patients underwent routine rehabilitation. Those in the MT, CCFES, MT combined with CCFES, and control groups received MT, CCFES, MT combined with CCFES, and routine rehabilitation alone, respectively. Upper limb motor function, activities of daily living, and corticospinal excitability were evaluated before and after a 3-week intervention period. RESULTS: MT combined with CCFES demonstrated a significantly greater therapeutic effect on motor function of the paretic wrist than CCFES, MT, or routine rehabilitation alone. However, there was no significant difference in the overall motor function of the affected upper limb, activities of daily life, or corticospinal excitability between the MT combined with CCFES group and the other three groups. CONCLUSION: MT combined with CCFES may be a potential adjuvant therapy to promote motor function in paretic wrist after stroke.


The combined therapy of mirror therapy (MT) and contralaterally controlled functional electrical stimulation (CCFES) may be a potential adjuvant therapy to promote motor function in paretic wrists after stroke.MT combined with CCFES could be incorporated into rehabilitation programs for post-stroke patients with impaired upper limb motor function.Exploring the potential benefits of combining different rehabilitation therapies could inform the development of more effective interventions.

As one of the main methods for sludge treatment, recovery of renewable biogas energy by anaerobic digestion (AD) is a promising strategy to deal with the conflict between carbon neutralization and sharply increase of sewage sludge. Humic acid (HA) in sludge is a major inhibitor of biogas yields and needs to be removed or pretreated. However, as the graphene oxide-like material, HA is an ideal precursor for the preparation of energy storage materials with high performance. Based on that, this study i) proposes the extraction and utilization of HA in sludge, ii) discusses the feasibility of HA-based materials after thermal reduction as electrodes for supercapacitor, and iii) investigates the factors with positive influences on the structure and electrochemical performance. It reveals that, with a synergistic effect of purification and activation at a low mass ratio, the HA-based material exhibits superior capacitive performance with the highest specific capacitance of 186.7 F/g (at 0.05 A/g), as well as excellent rate capability and cycling stability. Sludge is verified a cheaper and more abundant precursor resource of HA for energy storage application. The results of this study are expected to provide a new green, energy-efficiency and sustainability way for sludge treatment, which has the double benefits: efficient conversion and capture of bio-energy during AD process, and high value-added utilization of HA for supercapacitor.

Patients with germline neurofibromatosis type 1 (NF1) microdeletions frequently exhibit hereditary syndromes such as cardiovascular anomalies and have an increased risk of malignant peripheral nerve sheath tumors (MPNSTs). This study aimed to identify the genes codeleted with SUZ12 that are related to MPNST. We used differential gene expression and enrichment analyses to analyze the SUZ12-mutant and SUZ12-wild-type gene expression profiles in the GSE118186 and GSE66743 datasets in Gene Expression Omnibus (GEO). PPI network analysis combined with MPNST patient survival analysis was used to identify ADCY1, which catalyzes the conversion of ATP to cAMP, as a key gene. Moreover, chromatin immunoprecipitation sequencing (ChIP-Seq) showed that the distribution of H3K27me3 in the ADCY1 promoter region and gene body was significantly reduced in SUZ12-mutant cells. To verify the role of ADCY1 in SUZ12 mutation, we used RNA interference and plasmid transfection to interfere with SUZ12 expression in plexiform neurofibroma (pNF) and MPNST cell lines and then treated the cells with forskolin, IBMX and H89. ERK phosphorylation was accelerated and prolonged after siRNA transfection, especially in ipNF05.5 cells, and the intensity and duration of ERK activation were reduced after SUZ12 overexpression. Importantly, the level of p-ERK was consistent with that of Rap1-GTP. Moreover, H89 completely blocked Rap1 activation and the changes in the p-ERK level after SUZ12 siRNA transfection. In conclusion, our findings suggested that SUZ12 loss potentiates the effects of NF1 mutations by amplifying Ras signaling through the ADCY1/cAMP/Rap1/ERK pathway and that SUZ12 may serve as a therapeutic and prognostic biomarker in NF1-associated neurofibromas.

Chronic pain and sleep disturbance are highly comorbid disorders, which leads to barriers to treatment and significant healthcare costs. Understanding the underlying genetic and neural mechanisms of the interplay between sleep disturbance and chronic pain is likely to lead to better treatment. In this study, we combined 1206 participants with phenotype data, resting-state functional magnetic resonance imaging (rfMRI) data and genotype data from the Human Connectome Project and two large sample size genome-wide association studies (GWASs) summary data from published studies to identify the genetic and neural bases for the association between pain and sleep disturbance. Pittsburgh sleep quality index (PSQI) score was used for sleep disturbance, pain intensity was measured by Pain Intensity Survey. The result showed chronic pain was significantly correlated with sleep disturbance (r = 0.171, p-value < 0.001). Their genetic correlation was rg = 0.598 using linkage disequilibrium (LD) score regression analysis. Polygenic score (PGS) association analysis showed PGS of chronic pain was significantly associated with sleep and vice versa. Nine shared functional connectivity (FCs) were identified involving prefrontal cortex, temporal cortex, precentral/postcentral cortex, anterior cingulate cortex, fusiform gyrus and hippocampus. All these FCs mediated the effect of sleep disturbance on pain and seven FCs mediated the effect of pain on sleep disturbance. The chronic pain PGS was positively associated with the FC between middle temporal gyrus and hippocampus, which further mediated the effect of chronic pain PGS on PSQI score. Mendelian randomization analysis implied a possible causal relationship from chronic pain to sleep disturbance was stronger than that of sleep disturbance to chronic pain. The results provided genetic and neural evidence for the association between pain and sleep disturbance, which may inform future treatment approaches for comorbid chronic pain states and sleep disturbance.

Drought is a complex natural disaster phenomenon. It is of great significance to analyze the occurrence and development of drought events for drought prevention. In this study, two drought characteristic variables (the drought duration and severity) were extracted by using the Theory of Runs based on four drought indexes (i.e., the percentage of precipitation anomaly, the standardized precipitation index, the standardized precipitation evapotranspiration index and the improved comprehensive meteorological drought index). The joint distribution model of drought characteristic variables was built based on four types of Archimedean copulas. The joint cumulative probability and the joint return period of drought events were analyzed and the relationship between the drought characteristics and the actual crop drought reduction area was also studied. The results showed that: (1) The area of the slight drought and the extreme drought were both the zonal increasing distribution from northeast to southwest in Yunnan Province from 1960 to 2015. The area of the high frequency middle drought was mainly distributed in Huize and Zhanyi in Northeast Yunnan, Kunming in Central Yunnan and some areas of Southwest Yunnan, whereas the severe drought was mainly occurred in Deqin, Gongshan and Zhongdian in Northwest Yunnan; (2) The drought duration and severity were fitted the Weibull and Gamma distribution, respectively and the Frank copula function was the optimal joint distribution function. The Drought events were mostly short duration and high severity, long duration and low severity and short duration and low severity. The joint cumulative probability and joint return period were increased with the increase of drought duration and severity; (3) The error range between the theoretical return period and the actual was 0.1-0.4 a. The year of the agricultural disaster can be accurately reflected by the combined return period in Yunnan Province. The research can provide guidelines for the agricultural management in the drought area.
